Atorvastatin calcium (ATR) is mainly used for lowering blood cholesterol, while
ezetimibe (EZ) is used for the treatment of hypercholesterolemia and homozygous
sitosterolemia. Combination of ATR and EZ has the advantages of greater
therapeutic effects than with either drug alone. This combination causes
manifold reduction in LDL cholesterol level as compared to double dose of the
individual drug when used alone. Moreover, using of EZ with ATR enhances the
effects of statins at lower dose and reduces the associated side effects. The
UV-vis spectra for ATR (solid curve) and EZ (dotted curve) are shown below.
(1) Why is it advantageous of using a cmobination of the two drugs over
individual?
(2) Suggest a spectrometric method for the determination of the contents of the
two drugs in a pure mixture containing the two drugs.
(3) Is sample pretreatment necessary for the determination of the
concentrations of the two drugs in blood samples prior to the spectrometric
analysis? Why?
